Brief Introduction:
Peptone is a mixture produced by partial hydrolysis of proteins, containing free amino acids, oligopeptides and other protein components that are easily digested by microorganisms. Its preparation process usually involves enzymatic or acidic hydrolysis, and it is widely used as a nitrogen source and sometimes as a carbon source in microbial culture media. Studies have shown that the quality and yield of peptone can be improved by optimizing enzyme concentration, pH value and temperature using different sources of protein (such as dark meat of yellowfin tuna and soybeans) and waste (such as fish processing waste). For example, the optimal conditions for hydrolysis of soybeans using papain are 0.3% enzyme concentration, pH 5-7 and temperature 40-60°C, while high concentrations of protein hydrolysates can also be obtained when treating fish waste using alkaline and acidic hydrolysis methods. Peptone is used as a stabilizer in culture media due to its buffering capacity. Experimental studies have shown that peptone can effectively support the growth of a variety of bacteria (such as Escherichia coli, Bacillus subtilis, etc.), demonstrating its potential for application in microbial culture.
